・Importance of DNA Template Design
Our recent research has shown that DNA sequences have a significant impact on the yield of protein synthesis. Even with the same amino acid sequence, variations in DNA sequences can lead to different levels of protein production. This effect is independent of biological distinctions such as prokaryotic or eukaryotic. Certain DNA sequences tend to produce higher or lower yields of protein, and these trends have become increasingly apparent. For more details, see 6 Tips about the template DNA Design.
Considering these critical factors, DNA template design can often be a challenging and complex task.
To address this, we have developed "PUREcodeTM", an automated design software that incorporates these factors to easily optimize DNA sequences. This tool simplifies the design process, and enable to access it easier.
・How to Get Started
1. Submit your amino acid sequence: Access the form to enter your amino acid sequence.
2. Receive a custom DNA design: Within 1-2 business days, you will receive a text file containing a DNA sequence optimized for PUREfrex.
・Recommended Next Steps for DNA Synthesis
To confirm whether your protein can be synthesized with PUREfrex®, we offer an affordable kit called PUREfrex® 2.0 mini.
This kit contains a universal primer designed for optimal use with PUREfrex®. Our research has shown that the 5' UTR sequence of the template DNA can significantly affect the yield of protein synthesis. For more information, please see this poster.
PUREfrex® 2.0 mini contains a universal primer with a 5' UTR sequence optimized specifically for PUREfrex®. Since PUREfrex® has low contamination levels of RNase and other degrading enzymes, you can use PCR-generated template DNA.

PUREcodeTM, our DNA sequence design software, can be installed directly on your PC. Please note that there are specific system requirements, so we recommend that you contact us for further details to ensure compatibility. The software is distributed free of charge.
