The efficiency of translation on the ribosome is affected by the nucleotide and amino acid sequences of the synthesized proteins. In particular, the nucleotide and amino acid sequence in the N-terminal region significantly affects translational efficiency.
In this study, we investigated the effect of the N-terminal sequence on protein synthesis in the PURE system (Product name: PUREfrex®). We found that a single amino acid substitution in the N-terminal region significantly altered the translation efficiency in the PURE system, especially at low temperatures, which gives us useful suggestions for the expression of the protein of interest in vitro and in vivo.
